I can't confirm it since I didn't use sub for the final form, but apparently her final form has ASW high enough to sink your sub.
I used a sub for all forms and she didn't have ASW herself. Neither did the escort. What happened was that the transports changed into late model DDs and sank my sub (with a damecon on it, of course).

I'm pretty sure that her last form proced the DameCon on my Yuu. E: Just checked my Screenshot folder. Either her final form or that flying fortress did it.

This may be the first time so far that I've ever prayed for my battleships to take out the DDs first ^^;;;;
Didn't have Yuu or boilers or high level subs, so I used KTKM instead. About 20 runs and finally got through. Absolute pain. It didn't help that my Suzuya and Kumano were underleveled as hell. In retrospect, really should have chosen a difficulty other than hard.

I don't even need to equip any damecon on E-5 (put torpedo and WG42 on my Ro chan) and yeah, enemies fleets can't sink my Ro because the BB just decimated every enemies DD before they can even shoot. Flagship CL usually damage Ro to medium at the Night battle node, but it's just that.
Anchorage Water Demon still hits REALLY hard at night tho.
I used Shioi, and she only sank three with a damecon, and it was first the DD (had a tough run before the night battle node so she sank during it, and the second time was the new DD that was at the boss final form). The boss did sink Shioi once at final form, but well, Shioi was already at 1hp at the beginning so can't confirm if she had any ASW boost.

Seriously, even with damecon on, those battles were bad for the heart. I had an acquaintance on twitter who did have his heart jumping when he saw that happening with his Ro-chan. It never happened to me but that shit left me sweating cold. I trried two runs without a sub and I can tell you that it's a bad idea.

Imagine you lost your damecon on a previous battle and didn't give her a new one and then... you realize it and go back to the base with your tail between legs.
Yep, you'll feel like like you lost a couple of years if that happens.
Ah, the Anchorage Demon, it took me quite a while to beat her... mostly because my Ryuujou was kinda of underleveled (level 61), so she got the shaft the most on my E-5 run.
Tried to brute force my way with two carriers to get air superiority and KTKM for a pre-emptive torpedo, but failed to kill the demon during the night battle phase. Seriously, unless your submarines are severely underleveled, it's not worth it because of the pre-boss night battle node and both Anchorage and Flagship Ru shelling can deal massive amounts of damage.
The Anchorage and Floating Fortresses torpedo bombers they caused me quite some grief, Choukai got rekt twice in a row. Tried using AA cut-in setups on my FBBs, but then they didn't had enough punch to kill the Flagship Ru or the Anchorage (even with support expedition).
In the end, I've used Iku to tank, since she has the highest attack power amongst subs (she managed to sink a few DDs and transports with pre-emptive torpedos), equipped Kongou and Kirishima with both AP and Type 3 shells, and sent support expedition.
Kirishima killed the Anchorage at night with a double attack: 254 and 294 damage. Yeah, I've had to mix shells to kill that demon reliably.
Long story short, E-5 was a pain in the butt and now, if you excuse me, I'm gonna have some therapy and a shot of vodka.
