I'd say it depends if he is wearing armor or not, otherwise he won't really have anything to defend against time stopped knives.
But then again, he still has plenty of weapons that could also 1-shot Dio, so considering both's egos, it's a battle of whoever quits playing around and takes it seriously first.

let me disagree, while dio has an ego comparable to gil's. He is by far one of the most calculating villains I have seem. While jojo has always been a bit dependant on deus ex machinas, I strongly believe that dio had should have won. I mean, passing from being able to move in stoped time for 1~2 seconds to stop time so suddenly, its no wonder why he lost. We're talking about the man who after impaling his enemy with at least 5 knives in fatal areas decided to shoot him, then check that he wasnt breathing nor his hearth was beating, AND after that decided to decapitate him just to be sure
