Chances are that Yukari will 1) Runs away crying like a hurt highschooler (Just as Trolled) 2) RAGE (it would still be Just as Trolled anyway).
Some may call it a low blow, but considering how much abuse Remi gets in this comics, i think it would be fair for her to hit someone with a low and hard blow at least once.
Xelmitsu, that's even better and as factual as Yukari's situation with Rinnosuke.
Actually, let's combine both, i bet it would be a harder blow would and it would be priceless to see what kind of reaction Yukari gets for being reminded about her failure at hitting on both the trademark Miko and the Weird Shop Owner.

一つ is normally used for a number of things ("one"), not for the order of things ("first"), though.
For the whole bubble, I'm getting something along the lines of, "As you are a master yourself, you have to take responsibility for the things you have said." The italic text indicating the part I am uncertain of. Anyone else have thoughts to share on this?

Knowing Yukari, even if Remilia said all that about her and Rinnosuke, she would probably as calm as she is right now and will counter with a even worse comment. She's one of them super trolls that can take whatever you say calmly and reflect them back at 5x the strength and pain.
一つの主である以上 means something to the effect of 'as a master yourself, you...'. To break it down, "一つの主である" is 'as a master', and the '以上' adds 'more important than the fact that'. My explanation isn't very good, this is tough to put into English. Anyway, I hope my line is satisfactory, because it conveys the relevant meaning.
