If you did but missed it, peni briefly reappears amongst the spider people, with a different appearance since the first movie being less happy and more sullen since joining the spider society. She's among the spider people who're trying to convince Miles to let his father die to save his dimension (but she doesn't say anything and later defects), hence why she's saying Miles in a more sorrowful way.

Isn't it also a nod towards Peni's original design from the comic she appeared in? (e.g. post #3277120, post #5915700) The cutesy bouncy anime version of her was more a re-design for the first Spider-Verse movie. In the original comic it's pretty serious and she's a pretty somber teen.

Poeple theorise that she's undergone her "cannon event" since the first movie, with people pointing to her baggy eyes and overall tired state, her cannon event being that her only friend, aunt and uncle who were the only people close to her all die. Also makes sense considering her universe/comic takes inspiration of Neon Genesis Evangelions as well.
